LEUCOLIEVA 300 mg contains Leucovorin Calcium (Folinic Acid), a biologically active reduced form of folic acid. It is used to protect normal cells from methotrexate toxicity (rescue therapy) and to enhance the antitumor activity of 5-Fluorouracil (5-FU) in colorectal and other gastrointestinal cancers.
Leucovorin Calcium is indicated for:
1. Methotrexate Rescue Therapy
– After high-dose methotrexate to reduce toxicity
2. Combination with 5-Fluorouracil (5-FU)
– Enhances efficacy in colorectal cancer regimens (e.g., FOLFOX, FOLFIRI)
3. Accidental Methotrexate Overdose
4. Megaloblastic Anemia due to Folate Deficiency
– When oral therapy is not suitable
Common side effects:
• Nausea
• Vomiting
• Mild allergic reactions (rare)
Serious side effects:
• Hypersensitivity reactions (very rare)
• Seizures (rare, at very high doses)
Generally well tolerated.
Dosage depends on indication, chemotherapy protocol, and methotrexate levels.
Typical dosing:
• Methotrexate rescue:
– 10–25 mg every 6 hours, adjusted based on MTX plasma levels
• With 5-FU:
– 100–400 mg/m² IV, as per protocol
